A compiler bug exists in the current ABI model 430A peptide synthesizer when running NMP Fmoc cycles. Following a double couple cycle, the activator cycle fails to begin until the previous reaction vessel cycle is completed. This error adds about 15 to 52 minutes to each subsequent cycle by increasing the waiting times, but does not appear to have a significant adverse effect on the quality of the peptide synthesized. The longer wait times will continue to be present in the following syntheses.
We have been able to rectify the problem only by resetting the instrument either prior to the next synthesis or during the current synthesis. We would be interested in knowing if anyone has found a better solution to this problem.
