PROTEIN SEQUENCE COMMITTEE


The Protein Sequence Research Committee has prepared and characterized the sample for this year, ABRF-94SEQ, which will be mailed in mid-February to all facility directors with current ABRF membership. The emphasis of this year's study is the identification of cysteine and tryptophan residues in a protein. Based on past ABRF studies, the ability of member laboratories to identify these residues has emerged as a consistent problem. The goals of this year's study are to provide current Cys and Trp identification methods and to identify methods that are most reliable and readily adaptable to routine use in facilities. Included with the sample are methods for the reduction and derivatization of cysteine and a separation method for PTH-Trp on ABI sequencing systems. Two aliquots of ABRF-94SEQ are being mailed to allow analysis by more than one method, if the director so chooses. Please make every effort to analyze the sample, return the chromatograms (including standards), data reporting sheet and short questionnaire using the return mailing label.
