PEPTIDE SYNTHESIS RESEARCH COMMITTEE

For 1996, the Peptide Synthesis Research Committee will conduct a study to examine racemization during peptide assembly. Participating laboratories will be asked to synthesize a short peptide with two amino acids susceptible to racemization. The peptide can be synthesized using either standard (hydroxymethylphenoxy) or chlorotrityl resins. The purified peptides submitted will be analyzed by the committee using the following methods: amino acid analysis, MALDI-TOF and ESI mass spectrometry, chiral-HPLC, reverse phase-HPLC, polarimetry, and enzymatic digestion procedures. The results should provide insight into the extent to which racemization needs to be monitored in core laboratories.
