Dear Colleagues,
I have bought recently a Kromasil 250x4.6mm C4 HPLC column from Supelco. I
planned to use this column in standard water/TFA/acetonitrile buffer system
but it appeared impossible: always when I try to run a standard gradient
from 0 to 80 % of acetonitrile the broad big (2 AU) peak appears at about
40 %. This peak is equal at 220 and 280 nm and is absent when I mount C18
or C8 columns. I flushed this new column with pure acetonitrile and water
but without any effect. Is in my acting something wrong or the column may
be bad ? Any ideas will be greatly appreciated !
