Good Day,

We are having a problem with our DigestPro instrument. For those of you
out there in ABRFland that have this instrument, I'd appreciate your
feedback.

The problem is this. During the addition of the reagents by the robot, the
liquid spills out of the tops of the reaction tubes. Not all of the liquid
goes in the tubes and much of it stays at the top. This happens regardless
of the gel percentage. We also cut the gel band into 4 to 6 1mm3 chunks
(no crushing). So, I do not think that plugging of the hole in the bottom
is the issue. What happens is that the acetonitrile used in the digest
reaction destroys the plastic polycarbonate lid when the liquid spills over
the tops of the tubes. We replaced the polycarbonate lid included with
the exchange kit for the DigestPro so that the tubes without adaptors can
be used. This problem has already begun to happen with the new lid!! Of
much greater concern is the inconsistency of liquid delivery during the
washing and reduction/alkylation steps. This probably relates to why some
digests work and others, mysteriously, do not. Any advice would be greatly
appreciated.
