When we first used dRhodamines, we experienced very low signal, but solved
this by bumping up the gain on the instrument to 4. That took care of it for
us. Big dyes are certainly worth it for most templates, but there are some
instances where you may prefer dRhods, e.g. di-nucleotide repeats, polyT
stretches, G/C rich sequence. We use Big Dyes exclusively with dGTP on some
of the sequences that show premature termination, about 1% as a rough guess.
