Dear Colleagues:
In recent discussions with PE Biosystems I came across a discrepancy in their
PNA Synthesis manual that I recently downloaded from their website. They list
the Activation and Coupling time as 8.25 minutes (Figure 1-4, Ch. 1).
However, in discussing this with their technical support, they said that their
standard protocol is to activate and couple for at least 16 minutes. What
coupling times, and for that matter, what cycle times do you use routinely for
those of you who synthesize PNAs? I will be interested in learning more about
this. Also, what is your average step-wise coupling efficiency for 2 umol PNA
synthesis?
