We recently digested a gel sample that was brought to us by a researcher which was stained with a colloidal silver stain. There seems to be a large amount of signal suppression, on Maldi, from the stain as evidenced by some peaks showing up in the blank and no peaks and a high baseline signal from the sample gel slice. I remember seeing several posters at past ABRF meetings dealing with Farmer's reducer to process samples with this type of stain. We have tried to cleanup the samples after digestion with Zip-tips without any success. My questions are:
1.) What are the references for this procedure?
2.) Will it work on the sample after digestion?
3.) What are the best references dealing with stains to avoid in gel digests destined for MALDI?
