I wish to thank everyone for helping me with my histidine problem especially
Anita Everson, Fperini, Steven Johnson and Igor Rodionov. I did not want to
post until I could provide an update and here it is: The yield and purity
have been dramatically improved by both increasing the length of coupling
AND extensive washing of the resin following HBTU mediated coupling. I am
also using HOBt in the presence of His but introducing it as a solution with
HBTU and increasing the coupling time from 30 minutes to 3 hours. The yield
has tripled and the boss is happy. Thanks again.
