We have been running a DNA facility for a few years now and use a 377
machine with 48cm plates. We used to get good data, with reads of
700-800 bp from good samples. Gradually over time though, I have
noticed a drop off in read length and sample strength. Even with our
pGEM standard we are noticing that the traces are very feint (although
we use the same concentration and conditions as always) and
customer samples are often too weak to read, so I have to repeat them
with an increased number of thermal cycles.
As far as I am aware, none of the conditions, chemicals etc have
changed. We use a glass washer and never get any dirty plates. Could
there be any residue on the plates that effect the runs, but not show up
on the gel image?
