Long time ago, phthalic anydride was used. The reason for this was to produce
a number of
acidic peptides. This was in the days of classical(not solid phase) peptide
synthesis and
ion exchange purification as a first step.Phthaloylation was also done in
solid phase
synthesis.I tried the procedure a number of times, and results were not too
good.And,hardly
anyone uses capping in Fmoc chemistry, when it should be used..

Subject: Pepsyn: capping strategies

Has anyone had any luck using something besides acetic anhydride to cap
the unreacted amines? Perhaps something hydrophobic that will change the
retention time significantly. Thanks in advance...
