Hi Trudy and Terry

The Pharmacia Spin columns work very well indeed for desalting small
quantities of oligos. For larger ones the NAP 5,10, 25 should be better. The
protocol is very easy. Have a look on the Pharmacia catalogue. They tell you
which one you have to use depending on your proper application.
You cannot compare the purification done with an OPC cartdrige and sephadex.
In the first case (and if you run a DMTON synthesis) you can separate some
failures sequences from the expected DMTOn compound and depending on the
further elution step you can desalt the oligo. With sephadex most of the
failure sequences (except the very short ones < 8 mers) go throw the column
and coelute with the expected compound. But the desalting step is very
efficient since all the small molecules are trapped inside the resin bed .

Biorad sells as well desalting columns for oligo (P2...if I remember)
