ABI3700

Hi,

We recently had our ABI3700 installed. So far, we have performed about 12
runs. The first run came out very nice, with long reads and clean
sequences, however, I noticed that there were spikes in the chromatogram
files for six of the samples. The second run showed a higher frequency of
spikes and the situation continued to worsen. We decided that the problem
was due to air bubbles in the polymer and therefore flushed out all of the
polymer and water and replumbed the system. Also the cap to the polymer
bottle was replaced to rule out improper venting. However, while the machine
was in mid-run we noticed that new bubbles had formed in the polymer bottle
and were being pulled into the system tubing. Has anybody seen this problem
and if so how did you resolve it?
