I used to do synthesis on a VEGA machine which had no feedback
monitoring. I think they now exist only in the graveyards of
technical equipment . However the only way I could monitor a synthesis
would be to do the trusted Ninhydrin test at every step, or every
crucial step ( depending on the sequence). With fmoc synthesis however
you might get false positives when testing a coupling, because the
pyridine in test solution 2, might cleave the fmoc off of the resin.
You will have to use a different reagent to test iminoacids like Pro
( isatin solution works). Hope this helps.
