Alex,

An alternative to AAA for protein/peptide quantification is total
Nitrogen determination, eg, by Kjeldal. You need to know % N or AA
composition of your peptide and you need much more material. In the
past I have successfully used Kjeldal analysis to confirm the
accuracy of results obtained by AAA on recombinant proteins. I think
there are now better methods/instruments for doing Total N which may
be worth looking into, but generally AAA is preferred because of its
sensitivity and it also gives an AA composition.
