FW: Digest - anomalous Glu-C cleavage between SER & GLY

The unusual cleavages that I got were between glycine and glutamic acid and
between alanine and glutamic acids. I have not seen a cleavage between
between ser and glycine. I also was using ammonium bicarbonate buffer at a
pH of 7.5. I am curious to know what was the enzyme substrate ratio. I
noticed that unusual cleavages by endopeptidases were quite common when the
enzyme concentration was 1:10 or higher.

Subject: Digest - anomalous Glu-C cleavage between SER & GLY

Does anybody have experience of endo Glu-C digestions in ammonium
bicarbonate
pH 8.0 cleaving between SER and GLY residues? More completely the sequence
is
YLRSGGKP. Any clues would be welcome.

Thanks

Mike
