Susan,
my guess is that the purity of the lambda template is a point. lambda is
not easy to get very pure unless one uses columns. very frequently people
are having trouble even in cutting it with enzymes.
with respect to the universal primer.. of course you did not use any M13
universal primer, did you? (since lambda doesnt have those binding sites)
