Oligonucleotides

Has anyone been able to detect depurination in synthesized oligonucleotides
using CE?
I have recently been seeing significant n+1 shoulder peaks on longer oligo
syntheses and am looking for possible explanations. I am synthesizing using
standard phosphoramidite chemistry with 0.25 M DCI and 3% TCA as the
activator and deblock reagents, respectively.
I am aware that TCA can cause significant depurination with increased
exposure times and am wondering if this could be an explanation to the n+1
shoulder that I am seeing.
Are there other explanations?

Thank you,

Aaron Schohn
