Dear ABRF
                I have just tried to attach rhodamine to the N-terminus of a
peptide using Rho-ITC (Sigma product R1755) with a beta-alanine spacer. The
reaction all but failed...I can detect maybe 2-5% product in the crude by
mass spec. Does anyone routinely attach Rhodamine synthetically and covently
to peptides. Do you use the same isocyanate reagent and procedure which
works so well for fluorescein? Does the expensive NHS reagent from Molecular
Probes work better? The Sigma product R1755 is supplied as the Cl- salt (not
the HCl salt). Does this require special handling to activate it successfully?
