Two tips from our local 2-Der who's been getting up to speed -
1. replacing bis with PDA (piperazine dicarylamide, new product,
can get from Biorad) causes a significant reduction in background,
and
2. this method from Swiss-Prot at
http://www.expasy.ch/ch2d/protocols/protocols.fm4.html
is apparently excellent, although it is a long one.
