As to your problem of gelatin-containing samples to be prepared for
MALDI-MS:

I think you have to get rid of the gelatine ! Your sample won't crystallise
and, moreover, gelatine (highly X-linked collagen-like proteins) may give
you background signals. You want to analyse proteins/peptides, am I right ?

You can try a simple RP-purification (SepPak (Waters) or a micro-column,
depending on your sample amount). Gelatine will stick to the column whereas
you can elute your peptide/protein sample with a moderate amount of ACN. I
case of very little sample, you can prepare tiny columns in a GelLoader tip
and directly elute with matrix.
If you absolutely want to get around a small-scale purification step, you
could also try a sample prep with nitrocellulose (sandwich method).
You may need to heat you sample solution a bit, since gelatine is soluble
in warm water but becomes viscous when cold.

Has anybody ever tried to get mass spec data on a sample that contains
Gelatin? We are using a Bruker Reflex III MALDI mass spec. When I use
Synapinic Acid with the sample, it turns yellow and becomes very viscous.
It appears that I am not getting any crystals. Any suggestions?
