Mary,
We recently purchased the Hitachi L-8800 and have found the
instrument to be very good. We have found that the L-8800 produces very
reproducable results. Recently we had to reanalyze a protein sample which
was stored frozen for about a month. The Hitachi system produced nearly
identical results compared to the first time the sample was analyzed. There
were over 100 injections on the same column between analysis. We have also
found the instrument to be relatively easy to learn. At this point there is
a team of people at Hitachi ready to help if you have problems.
There are some things that you will find different from the Beckman
instrument. We have found with Hitachi's buffers and gradient that the
internal control Norleucine overlaps Tyrosine. Norvaline was substituted
successfully instead. The column is claimed to last for over 1000
injections. At ~600 injections, we have noticed peak tailing and it is not
likely to make it to the 1000 mark. Keep in mind that these columns are
very expensive. Hitachi is supposed to be offering a trade-in for the old
column but has yet to implement this option. I'm told this will be soon.
Overall we are very pleased with the instrument and would recomend
it as a replacement for the Beckman 6300.

We are currently using a very old and fast ailing Beckman 6300 amino acid
analyzer and would like to replace this system with a Hitachi L-8800 amino
acid analyzer. We would like to know what other lab's experiences have been
with the Hitachi system. We would prefer the standard ninhydrin method. We
appreciate any feedback. Thanks!
