Hello all;
Just writing to share the latest breakthroughs in solving problems with my
473 protein sequencer. In a nutshell, the hydrophobic residues were
disappearing from my blotted samples. There were tiny leu and ile peaks but
valine was nonexistant. Lysine also nonexistant. PTH standards were fine.
Many thanks to Bob Apfelzweig at ABI for looking at my chromatograms and
coming up with some excellent suggestions, one of which was to leaktest
(something I had neglected to do after installing a new valve block #1).
There proved to be a bad leak at the bushing on the line going from the
valve block to the cartridge. Fixed it and got fine results on a BSA run
last night. I don't understand why a leak would produce this kind of effect
but I'm VERY happy to be up and running again.
