Hi! Folks:
I probably have the following sequence:
KSCCPSTTARNIYXACRFAGGSRPTCASVSGCKIISGSTCDSGWNH,
X: N-(Hexosamine/Hexose)6
During the Edman sequencing, there is a blank in the 14th cycle, sugar-
linked N was traced out by the combination of AAA and similarity alignment
with known toxins. Hexosamine was also found during AAA.
My question is: are there any practical methods to qualify the sugars linked
in glycopeptides? We proposed the sugars by the mass difference between the
measured MS and the calc. mass of the non-sugar sequence. But there is still
a little big gap, not so accurate. And I guess I should also cleave the
sugar by Endo F or other enzymes to validate N14 and the sugars, where can I
find a related protocol? Which company supplies the better enzymes (let's
say, Endo F)?
Any comments are appreciated!
