You'll probably get lots of suggestions about IMAC tips and Ni/NTA agarose
beads for enriching for the peptide.
It's perhaps a bit antediluvian but the thing I'll mention is the volume by
Stewart and Young, "Solid Phase Peptide Synthesis" (probably in Biochem
library) in which they provide a recipe for a His-specific TLC stain, I
believe it was called the Pauly stain. This might allow you to survey (the
presumed earlier) HPLC fractions dotted to a PVDF strip. I'm assuming you
have some sample to spare - the sensitivity of a TLC spray will be lacking
relative to modern methods but at least you won't disturb/contaminate the
bulk of your fraction.
