Glycosylation decreases the efficiency of the automated Edman chemistry which often results in
rapidly escalating lags. Depending on what you know about the sequence and what you're trying to
find out, you may be able to live with that for short stretches of data. My experience with attempts
to get enough unambiguous sequence data to be used for cloning the genes of unknown brain proteins
(which often also required sequence that generated primers of not unreasonably high degeneracy) was
that deglycosylation was required.
