I will add my bit to the continuing discussion. As mentioned by
Marcus it is possible to identify amino acids substituted with a
monosaccharide using standard sequencing but the recovery is low. By
using a more polar solvent such as methanol to transfer to the
conversion flask better recovery of glycosylated amino acids
including larger oligosaccharides can be achieved. It is necessary,
however, to covalently attach the peptides/proteins to solid support
to use this method. Ref: Zachara and Gooley, Methods in Molecular
