I've been trying to do some in gel digestions with Glu-C, and I have had very little luck getting it to work. Usually I just see Glu-C autodigest products. Has anyone been successful in doing this routinely? I've tried phosphate buffer and ambic buffer at 37C overnight. Maybe I should try doing the digest at 25C? What concentration of Glu-C do people normally use for in gel digestions, of coomassie stained gel bands (I do not know the protein concentration)
Any help would be greatly appreciated
