Dear Jun,
you can have a look at the Phoretix software (which is the original to the
Pharmacia Imagemaster, this one is an OEM product) as well as on PDQuest
(BioRad) and on the software from Genomic solutions (which is in Germany
distributed by Perkin-Elmer [not PE Applied Biosystems!]). The latter one
runs on Unix systems while the two other ones are on Windows. To my
knowledge Phoretix does arrange demos of the software (but they have a CD
which will give you a presentation about the features) and PDQuest has a
demo version. Furthermore, to my knowledge, Melanie is meanwhile released
as version 3.
We are using the Pharmacia/Phoretix software and use a usual A4 scanner
with transparency unit. You should take care about choosing the scanner
because you have to be able to calibrate it, it must be a scanner which
moves the lamp together with the detector (otherwise you'll get systematic
distortions in the image, especially the intensity) and it should be
water-tight (for obvious practical reasons...). Pharamcia sells on, which
is a modified UMAX Powerlook 3 (which we use). It has a linear range from 0
to approx. 3.5-4 OD. Pharmacia has modified the attachment of the
transparency unit and the optics for a slightly different focussing plane.
They also made it water-tight by sealing. On the other hand the original
from UMAX works well in our lab. BioRad has one as well. You should
calculate at least something in the range of 1000 US$ for the scanner
because these are high end pre-press instruments which you can not compare
to those for the mass market which you can get for 100 $.
