Dear all,
Does someone have a protocol to make direct sequencing of 5' modified (for
example CY5 or CY5.5) oligonucleotides by Maldi-TOF using exonuclease
digestion kit like the Sequazyme?
Most oligonucleotides containing 5' modification are blocked against
digestion with BSP, so the protocol describe in the kit's notice doesn't
work for such oligonucleotides and the result is that in I can't obtain the
right sequence on the 5' end side.
