Lots of possibilities. Think "HPLC" and troubleshoot accordingly.
My first guess is that you went over-pressure, and the system shut itself
down automatically. Check the back-pressure on your system at 14%. If it's
high, try swapping in an old column, to begin to track down the plug. Other
places that 473s plug frequently include the sample injection valve and
associated sample loop.
I assume you're using a system that worked before, so the gradient command
set is probably OK. But you might check to make sure that you don't
inadvertently have a command for "0 ml/min flow rate" or "end analysis" at
the time for the start of the gradient.

I have a problem with running 473A protein sequencer. The
problem is: when I ran a STD cycle, HPLC pumps started initial 12%B
for 14 mins, but it stopped pumping right before Start Gradient step,
i.e Gradient won't start. Did anybody come across similar problem
before?
