Thanks to all who have responded. It sounds like I am not alone.
Answers to questions
1. The two columns I have tried are different lots
The first one was purchased in August S/N 1380 Lot #03G0-53696
(same lot as you Allison)
The second column was a replacement for this one shipped mid December
from tech support.
S/N 2108 Lot # 03M0-62919
2. To Carol --- I t seems I have tried a column made before and one after the
one you had troubles with. How is your replacement?
3. I use pretty much the same gradient as the original with the same 55 C temp
but I wil try the old one to see if it makes any difference.
4. The last suggestion from ABI is to run O/N interspersing stds and check to
see if they have improved with time. After about 75 injections the column is
the same and I have used a lot of chemicals.
As I work in a core facility where the researchers prep their own samples I
rarely have enough sample to repeat. and so need a column that works. Several
folks have sent alternate sources for columns. Which fit the ABI housings and
which work the best.?
Thanks again.
