We use 50-100 ng mouse DNA in 20 ul PCRs carried out in capped 96-well
plates with 0.2 ml well volumes. Do you use 5 ul PCR volumes in 384
plates? My initial concern using 5 ul volumes in 0.2 ml wells/tubes would
be possible adverse effects of volume changes (loss?) during thermocycling.
