I have recently started making peptides on a Rainin Symphony. I have
changed a few things - NMP instead of DMF, for instance. I will be adding
HOBt to the piperidine/NMP deprotection solution. Now I need some advice
from other Symphony users. Have any of you added HOBt to the
HBTU/N-methylmorpholine activation solution? Did it work? I have some
peptides to make that have a number of histidines, and I really worry
about racemization. I make up my activation solution fresh for each new
group of syntheses (8 at one time), so the solution won't be sitting on
the machine for any extended length of time - 2 or 3 days at most.
I would be extremely grateful for any information you can give me.
