Stephanie,

It could very possible remove a significant portion of the Fmoc protection.
Stay away from primary amines. I suggest using DIEA/DMF, as the
diisopropylethylamine does not participate aggressively in the
beta-elimination route for Fmoc removal.

Best regards, Dick

Subject: FMOC removal?

Dear All,

I would like to react FMOC-Cys-NH2 via the sulfhydryl group with an
N-bromoacetylated peptide to form a thioether. Will using TRIS as base (in
DMF) cause significant removal of the FMOC group? Thanking you in advance.
