Hello all,
We have run some colony PCR samples on a 377. The gel file appears to have
a very prominent
blue banding pattern with the remaining colors either non-existant or very
weak (a "washed
out" appearance). Has anyone seen something similar?

Investigator Sample type Purification
Result
1 colony PCR (600bp) Millipore plate blue
gel/"noisy &/or weak" chr.
2 colony PCR (600bp) Qiaquick blue
gel/"noisy &/or weak" chr.
3 Plasmid Qiagen mini prep gel
fine/seq. rxn. needs re-opt.
*Currently, I am using DYEnamic ET terminators ("mix")
*I am using a new lot of "mix".
*The plasmid samples and the control samples appear to fine on the gel
(color banding pattern),
however, the samples seem to need to be re-optimized.
*Investigator 1 and 2 said they have put colony PCR samples through
sequencing before with no
problems.
