Chuck,

In my experience, the majority of blocked antibody chains are blocked by
the presence of pyroglutamate. This can occur on either the heavy chain or
on the light chain. The sequence of the antibody chain can be easily
obtained by de-blocking with pyroglutamate aminopeptidase and subsequent
analysis by N-terminal sequence analysis. The procedure is relatively
simple and straight forward. The sequence obtained is equivalent to an N-1
sequence since the pyroglutamate is cleaved from the protein by the enzyme.
