Dear Janice, My note was a reply to Ron Niece's earlier query. I saw a peak
between P and Y and also before R. Ron saw peaks later with a much reduced
lysine yeild. The odd thing in Ron's case, (and in mine also if these are
indeed the same thing) is that they do not show up in every cycle with lysine.
I have occassionally seen the PTC lysine but generally when I was near to
overloading the sequencer.

Weather Inconsequential: A light dusting of snow here, with warm winter weather
(about 25-30 degrees). I hear that the bitter cold all you in the north have
been "enjoying" will be moving down our way soon.

Your recent message to the ABRF board has me a little confused. Where exactly on your
system are your K' and K" peaks eluting? I have a Procise like you do and I see a K' peak
eluting before P and after Y. It's usually 5-10% of the height of authentic K. I had
always assumed this peak was PTC-Lys and the "Modified PTH-Amino Acids in Protein Sequence
Analysis" book that Greg Grant put together indicates that PTC-Lys would elute in the same
location as my peak. The information in Greg's book on this peak came from an AB employee.
However, Dick Cook wrote me that PTC-Lys should not elute this early. The peptides which
I'm sequencing are all HPLC purified fragments of a synthetic peptide.

Weather: The artic air that froze the midwest is also affecting California, but of couse to
a much lesser extent. It may freeze tonight, although a band of showers is approaching
which will prevent the frost. It's overcast out right now (3:40 p.m. PST) and about 48
degs. (I'm guessing). Virtually everyone I talk to mentions how cold it is outside (or
inside, the heat in the building isn't working well). If mid-forties is "cold", I guess
there's one Californian (me!) who will get a frigid taste of winter reality in Baltimore
next month.
