I use control templates on both ends of the gel- for example on a 64
lane gel, I put controls in Lanes 1 and 64. A good control in the case
of cycle sequencing with Taq FS is 1.5 ul PGEM vector with 1.0ul
universal primer. I use the PGEM 3Zf(+) supplied by the Perkin-Elmer
Dye Terminator Cycle Sequencing Ready Reaction kits. The controls have
helped me at times when a large amount of empty lanes occur.
