Deb, As I recall, there an ABI written program that increases the cart. temp for
proline residues. The reg. lower temp. is used to lessen internal cleavages. I
just increase the temp. when I came to a known proline residues (I assume you
know where the prolines are).

Hi, all,
We are trying to get good N-terminal sequence data for the first 25
residues of a protein where there are 3 prolines (nicely spaced apart).
Has anyone had good luck with improved proline cleavage--I use an ABI 470A
protein sequencer with liquid phase.
