With regard to the issue of "fraction of reaction ratio" for BidDye terminator. I testes=d
2ul, 4ul, 6ul, and 8ul of the premix in a 20ul volume using both PGEM and our samples,
mitochondrial DNA (I brough up the volume to 20ul by adding dH2O). I got very good data when
4ul, 6ul and 8ul of premix was used. My later experience with this dye showed me that
removing excess dye is easier when 4ul of the premix is used in a total volume of 20ul. .
Other people may have had similar experience.
