I've recently attempted (successfully) to synthesize RNA oligos. None of
the phosphoramidites are available commercially so you need to synthesize
the monomers from carefully selected 13C labelled ribose and 15N and/or
13C bases, which tend to be very expensive. You can easily spend $4,000
just doing the glycosylation steps. Base, DMT protection and
phosphitylation require rigorously anhydrous conditions (I'm in Seattle!)
but prayer and molecular voodoo may yield enough phosphoramidite to label
one or two sites in your 27mer. I've seen price quotes for 250mg of 13C2
cytidine phosphoramidite at $30,000. If your investigator is still
interested have them contact me directly and I could provide more
information.
