Hello ABRF,
A collegue has asked me advice on how to purify a membrane protein to
send for mass sequencing. Although the sequence of this protein, Gas1,
is already known from DNA sequencing, it has a
glycosylphosphatidylinosito (GPI) anchor on an unknown residue, which he
wants to determine by the sequencing. His question, which I could not
answer and hope one of you can, was how he could purify the protein,
keeping it in solution, to a level, and in a final solvent, compatible
with mass sequencing. He has already partially purified it by ion
exchange chromatography, but in the presence of a detergent. Another
question was how to remove detergent from an eventually sufficiently
pure protein, while still keeping it in solution.
