I like a tryptic digest of myoglobin, which has peptides that separate well
by reverse phase. It has no cys, so you can buy myoglobin and digest it
without any manipulations. Digestion is quantitative with trypsin and you
can buy myoglobin that is quantitated already, so you know how many
picomoles you are starting with. It has several hydrophilic peptides that
allow you to judge whether you are getting good binding of small things.
And it has a couple that are reasonably hydrophobic and disappear when
things aren't right. The digested peptides are all very stable to freezing
and thawing, although there is some oxidation of some of them with time
(there is a tryp and some met).
I have also used a digest of cytochrome C as standard and gotten good
results--it also was stable to freezing and thawing and the peptides
separated well on most columns.
