Dear colleagues,
For more than 10 years I have been using a Pharmacia-LKB system=20
Alpha Plus 4151 for aminoacid analysis with IEC and OPA detecction: I=20
am very satisfied with the results we had with it. Now the instrument=20
is very old and we need to buy a new one. But the problem is the=20
money. The Pharmacia system is very expensive for that reason we=20
would like to consider the possibility of an rp-HPLC system.
But I have many questions.
I would like to know your general opinion about IEC versus rp-HPLC.=20
About columns: type, how long is the usefull life of the column, the=20
prices, the suppliers, if there is any possibility of regeneration of=20
the matrix.
About solvents: compositions, quality of the reagents to prepare them=20
by ourself, if there are some special considerations I must know.
About derivatization: which type is better? Fmoc, PITc or OPA.
What about the accuracy and reproducibility?
What do you recomend if we will do analyses of recombinant=20
proteins, some exotic aa=B4s and post-translational modifications on=20
the future instrument and also routine composition and concentration=20
determination on pure proteins?
Please share with me your experiences and give some advices,
