Joe asked to hear from those who have tried PVDF on the Procise HS. We
have one, and only one, sequencer. Our Procise HS was installed in
September of '96. From that time 46% of our runs have been blotted
samples (proteins ranging from 10K to 120K). Our peptides are usually
generated from digestion and HPLC separation and therefore run with
Biobrene on glass fiber filter. There is no problem shifting from GFF
to Prosorb in the Pulsed Liquid mode. Having retired an ABI 470, I am
a supporter of gas-phase sequencing, but running the Procise HS as a
multi-purpose instrument the PL cycles perform well (94% R.Y.).
I love the Procise HS but do not recommend it as an only sequencer.
The major reason being, it is much more expensive to run. The cost due
to columns (analytical and guard) alone is about $2.00 per cycle. This
is perhaps due to that fact that we must sequence every thing on it.
