Frank,
As far as your synthesis solution, your dark color is probably from the TRT
protecting groups on the Q and N, and the purple color is from TFA present
with Trp.
I would suggest putting a scavenger in your TIPS cleavage mixture - we use
Thioanisole on the Rainin when we cleave since the machine is in the lab and
the odor isn't as bad as EDT. The mixture we use is TFA-Phenol-Water-
Thioanisole-TIPS (82.5:5:5:2.5).
