Another reply to Peggy, tom et al. on the cLC:

On the 49X cLC's, I think several things are involved in the "wash
out." There's extra wash steps to reduce the bg and a pseudo-liquid
TFA delivery, thus more chances for wash outs. The latter should be
small, but you know how small things can be alot with micro
sequencing. Another factor is thermodegradation of the protein. With
the cLC cycles, the HPLC runs are longer than on the HT series (i.e.
the HT series have faster cycle times so less time is spent cooking in
the reaction cart.).

As a cLC owner, I'm very satisfied with the increased sensitivity,
even with the trade off in repetitive yields, thermodegradation, wash
out, higher maintenance, higher cost in reagents (they have to be
replaced more often), etc. Once it's running there not much
optimization to do when comparied to the old 47X series. I'm eagerly
awaiting a smaller column, 0.5 mm or less?

Dear Tom,

We didn't even consider the Procise; what is its sensitivity now? I
am a bit
late in this discussion, but I am also wondering about the washout on
the new ABI Procise HS capillary HPLC (.8mm column). Has anyone besides
Joseph Fernandez experienced this phenomena?
