It preferentially cleaves at the amino-terminus of aspartic acid but it also cleaves at the amino-terminus of glutamic acid, especially if the ratio of enzyme to substrate is less than 1:10, and in cases of overnight digest at 37 C.

Hi,
I have a protein that I digested with Asp-N (2% w/w for 24 hours). I
suspect that I am also getting some minor cleavage at the N-terminal
side of Glutamic Acid. Has anyone experienced this anomalous cleavage
when using Asp-N.
