Does anyone have a protocol, or know about one, for the in-gel digestion of
hydrophobic (membrane) proteins in SDS-PAGE gel pieces? I'm trying to
generate a peptide map of a seven-transmembrane receptor and I need a
sensitive (low picomole) method to do it. I will also be determining the
sites of post-translational modification. The resulting peptides will then
be analyzed by MALDI. Any suggestions as to which buffers, matrices, etc.
are best also would be helpful.
